This kit provides complete solutions for clients requiring a more accurate and robust assay system, or clients looking for high throughput capabilities in their toxicity testing regiment. The LumoPlate™ Ultimate Matrix Kit uses the kinetic method for determining toxicity. Matrix effects from sample coloration and turbidity are accounted for automatically and direct measurement of soil and sediment samples are conducted without time consuming correction procedures or filtration. Bacterial reagents are packaged to accommodate 1 entire 96-well plate, and ten plates are included in the kit. Cooling block and luminometer manufactured by Labrox are also offered at exceptional pricing to provide all required equipment that replaces more archaic methods. Individual replacement kit components can be purchased and reagent lease options are available for qualified clients based on volume purchases.

Contents:

1.   1243-705 – Freeze dried Aliivibrio fischeri reagent vials x 10 (1 vial can accomodate 1 full 96-well plate). Each kit will accomodate 10 full plates of analysis.

2.   1243-710 – 10 bottles of diluent (12 mL), sufficient to reconstitute all reagents.

3.   1243-715 – 1 bottle of Osmotic Adjustment Solutions (OAS) 25 mL.

4.   1243-720 – 1 bottle of sample diluent (500 mL).

5.   1243-725 – White sterile 96-well plates x 10 (sold separately, pricing available upon request)

6.   Labrox 96-well Plate luminometer for kinetic analysis according to ISO 21338 (sold separately, pricing available upon request)

7.   Cooling Block for 96-well plates and tube samples (sold separately, pricing available upon request)

The Kinetic Aliivibrio fischeri (formally Vibrio Fischeri) toxicity test Method was originally designed for solid samples is an improved method for determining toxicity data using luminescent bacteria according to ISO 21338:2010. Colour and turbidity of the sample are accounted and matrix effects can be largely eliminated. The method employs an automated luminometer capable of dispensing reagent and measuring luminescence at the same time and a microplate which permits up to 96 sample wells. Signal measurement immediately prior to reagent addition as well as continuously after reagent addition permits a fluid evaluation of changes in luminescence that automatically corrects for background effects. Initial signal intensity is compared to the signal intensity after the incubation period using a kinetic curve.

Most compounds of interest will produce a toxic effect immediately after contact with the bacteria is initiated results can be seen immediately. Depending on sample composition, the software can be employed to correct for interferences automatically in the calculation. Contact times can also be varied to observe effects of some chemicals that can only be seen after longer contact times.

This Aliivibrio fischeri (formally Vibrio Fischeri) toxicity test contains several benefits including
